GENERAL LABORATORY NOTES for IMMUNOGENETICS & TRANSPLANTATION


General Sample Requirements:

     Each blood tube or individual container must be labeled with:

          - Patient's legal full name

          - At least one of the following identifiers:

                    Medical Record Number   

                    Date of Birth

                    Social Security Number

                    HIC Number

                    Reference ID Number

          - Draw Date / Time

          - Phlebotomist signature

 

All samples must be maintained at room temperature.  Do not refrigerate.  All samples must be delivered to the lab as soon as possible.  Routine samples must reach the lab by 1500 on Fridays. 

 

 

***SPECIMEN REQUIREMENTS:

 

Yellow Top (ACD-A) Tubes (10 ml):

     - Avoid hemolysis.

     -  Keep at room temperature - Do not refrigerate.

     - Mix gently but thoroughly.

     - Tubes maybe obtained from ITL.  Call lab to request.

     - Samples for Cell Surface Markers must be received in lab on the day of collection.

 

LAVENDER TOP (EDTA) TUBES (3 ml):

     - Avoid hemolysis.

     - Keep at room temperature - Do not refrigerate.

     - Must be mixed immediately after blood drawing to prevent clotting.

     - Samples for Cell Surface Markers must be received in lab on the day of collection.

 

GREEN TOPS (Heparin):

     - Samples for Cell Surface Markers must be received in lab on the day of collection.

      The flow cytometric procedure for PNH evaluates both the erythrocyte and granulocyte populations for GPI-anchored protein expression.  CD15 and glycophorin-A, non-GPI-anchored proteins are used to positively idenitify granulocytes and erythrocytes respectively.  Our laboratory uses both CD59 and CD55 for analysis of GPI-anchored protein expression on erythrocytes, while CD16 and CD66b are used to evaluate the granulocytes.
