Absolute minimum amount: One slide
Method of collection: Collect serous exudate using physiological saline.
Transportation: Specimen must be brought to the laboratory within 15 minutes of collection.
Label instructions: Specimen must be labeled with patient name and Medical Record number.

Scheduling requirements : Must contact laboratory before scheduling this test, so that qualified personnel are available to examine the darkfield wetmount.
Method: N/A
Special Instructions: Serous fluid should be free of erythrocytes and tissue debris if possible. The lesion should be cleansed only if it is encrusted; use only physiologic saline (no bactericidal additives). May be necessary to gently abrade the lesion surface to yield a clear serous fluid. Transfer serous fluid to a clean microscope slide and cover with a coverslip (#1 coverslip is preferable to a #2). Ideal specimens for direct microscopic examination can be obtained if chancres, secondary lesions, or mucous patches are present. Do not send samples from oral lesions due to presence of many other spirochetes in the oral area that may resemble T. pallidum.
