Serous fluid should be free of
erythrocytes and tissue debris if possible. The lesion should be cleansed only
if it is encrusted; use only physiologic saline (no bactericidal additives). May
be necessary to gently abrade the lesion surface to yield a clear serous fluid.
Transfer serous fluid to a clean microscope slide and cover with a coverslip (#1
coverslip is preferable to a #2). Ideal specimens for direct microscopic
examination can be obtained if chancres, secondary lesions, or mucous patches
are present. Do not send samples from oral lesions due to presence of many other
spirochetes in the oral area that may resemble T. pallidum. |
